چکیده

SUMMARY Mice treated simultaneously with 1-0-D-arabinofuranosyl-cytosine (ara-C) and 6-thioguanine (6-TG) were protected from the lethal effects of 6-TG. No protection was afforded if ara-C was given 16 hr before the 6-TG. DNA synthesis in mouse bone marrow was immediately depressed to 3 to 12% of that of control in various experiments, after treatment with ara-C, 10 to 20 mg/kg; it then rebounded to 148 to 180% of control during the period 8 to 16 hr, and was below control at 24 hr. DNA synthesis in LI 210 cells treated in vivo with ara-C was depressed to less than 1% of control and did not begin to recover until almost 16 hr. However, after a second dose of ara-C at 24 hr, DNA synthesis in L1210 cells recovered much more rapidly. We studied mouse bone marrows and LI210 cells treated with 2'-deoxythioguanosine to determine the persistence of acid-soluble 6-TG nucleotides after a single dose. 6-TG nucleotides declined to low levels in 6 to 8 hr, exhibiting half-times of approximately 3 hr in both tissues.

Mechanisms of action of 6-thioguanine, 6-mercaptopurine, and 8-azaguanine.

The effects of 6-thioguanine on purine biosynthesis and cell viability have been examined in H.Ep. 2 cells grown in culture. Toxicity is not reversed by aminoimidazolecarboxamide, suggesting that inhibition of purine biosynthesis de novo is not the sole mechanism of toxicity. Also, 6-(methylmercapto)purine ribonucleoside, a potent inhibitor of purine biosynthesis de novo, produces more marked r...

Results of treatment of Ph'+ chronic myelogenous leukemia with an intensive treatment regimen (L-5 protocol).

Thirty-seven patients with Philadelphia-chromosone-positive (Ph'+) chronic myelogenous leukemia who were untreated or minimally pretreated were entered on the L-5 protocol. This protocol consisted of sequential treatment with splenic irradiation, splenectomy, arabinosylcytosine and 6-thioguanine, and L-asparaginase. Maintenance therapy was hydroxyurea or a multiple-drug regimen. The median surv...

Au Nanoparticle Loaded with 6-Thioguanine Anticancer Drug as a New Strategy for Drug Delivery

In this study we suggested a new strategy for drug delivery of 6-thioguanine (6-TG) as a cancer drug by loading of this thiolic drug at a surface of Au nanoparticles. For this goal, we synthesized Au nanoparticle (Au/NPs) by reduction of tetrachloroauric (III) acid solutions by sodium borohydride and characterized Au/NPs by X-ray powder diffraction (XRD), dynamic light scattering (DLS), Ultravi...

Hepatotoxicity associated with 6-methyl mercaptopurine formation during azathioprine and 6-mercaptopurine therapy does not occur on the short-term during 6-thioguanine therapy in IBD treatment.

BACKGROUND AND AIMS High concentrations of methylated thiopurine metabolites, such as 6-methyl mercaptopurine, are associated with hepatotoxicity during administration of the conventional thiopurines azathioprine or 6-mercaptopurine in IBD patients. Metabolization of the non-conventional thiopurine 6-thioguanine does not generate 6-methyl mercaptopurine. Hence, the aim of our study was to evalu...

ECCO consensus: evidence-based use of 6-thioguanine therapy in Crohn's disease?

With great interest, we have read the updated ECCO consensus on the treatment of Crohn's disease.1 Being highly interested in research on the clinical use of thiopurines, we feel that several additional commentsmay give opportunity to a more balanced, evidence-based conclusion on the discarded use of 6-thioguanine as an alternative thiopurine. Firstly, the authors state that 6-thioguanine is th...
